The best machine for running a web server is either Linux or OpenBSD running Apache web server with a MySQL database and PHP scripting language to access MySQL (Often called a LAMP server).
Currently 75% of all (true) web servers run Apache on Linux or BSD.
As for what type of hardware you need, like Andrew said, that would depend on how busy you think the server will be. As with all things computer, the better the machine the better the application will run. I would suggest at least a Pentium 4 grade machine with 80 Gig hard drive, and 1 gig of RAM...

How many concurrent connections do believe you'll have?
What are the minimal recommendations that your web server software provider suggests? (i.e. have a look at the apache documentation if you are using apache).
What other software are you going to need to run on the same server?
Is the application that you are serving high load?
etc.
Lots of questions that need to be answered before a recommendation I'm afraid.
You should also consider something like Xen or VMWare which allows you to run multiple OS sessions at the same time and can be swapped between machines very easily.
